Effects of N-ethyl-N-nitrosourea(ENU) on BALB/cAnNCrj male mice(1)

Abstract
We plan to carry out a systematic large scale development of phenotypic-driven mutant mice using the alkylating agent, N-ethyl-N-nitrosourea(ENU). We injected 200§·/§¸ ENU and 400§·/§¸ ENU intraperitoneally into BALB/cAnNCr j mice to decide the optimal dose of mutagenesis. After ENU injection, we investigated clinical signs and changes of body weight, lethality, temporary sterile period, fertility and breeding record with rearing program specially designed. Just after injected ENU, mice showed gloominess or/and drowsiness, but recovered in a couple of day. Body weight was reduced after every injection, but it was recovered to the original weight within 6~7 weeks after the final treatment in both doses. In 200§·/§¸ ENU group, the temporary sterile period was 15 weeks, average litter size was 1.8 heads, and average weaning rate was 60% from 17 to 23 weeks after ENU injection, but average litter size of control group was 5.8 heads and weaning rate was 97%. We could not have any breeding record in the 400§·/§¸ group, because there was no delivery by 23¢¥rd week.
